Assembling yeast complexes

he machine that is a cell is more than the sum of its protein parts. A large step toward understanding how those parts are so effectively put together has been taken by Patrick Aloy, Rob Russell (EMBL, Heidelberg, Germany), and colleagues. Their work has identified ‫ف‬ 100 yeast protein complexes and predicted interactions within and among many of them. " We were working on a large scale to mod...

Luminescent gels by self-assembling platinum complexes.

A platinum complex bearing a tetraethylene glycol chain has been designed and its self-assembly properties investigated. In solution, only a yellow phosphorescence of the aggregated species is observed. The complex gives luminescent gels of different colours with DCM and DMF, reaching up to 60% photoluminescence quantum yield.

Evaluation of a symmetry-based strategy for assembling protein complexes.

We evaluate a strategy for assembling proteins into large cage-like structures, based on the symmetry associated with the native protein's quaternary structure. Using a trimeric protein, KDPG aldolase, as a building block, two fusion proteins were designed that could assemble together upon mixing. The fusion proteins, designated A-(+) and A-(-), comprise the aldolase domain, a short, flexible s...

Optimized Affinity Capture of Yeast Protein Complexes.

Here, we describe an affinity isolation protocol. It uses cryomilled yeast cell powder for producing cell extracts and antibody-conjugated paramagnetic beads for affinity capture. Guidelines for determining the optimal extraction solvent composition are provided. Captured proteins are eluted in a denaturing solvent (sodium dodecyl sulfate polyacrylamide gel electrophoresis sample buffer) for ge...

An Essential Role for DYF-11/MIP-T3 in Assembling Functional Intraflagellar Transport Complexes

MIP-T3 is a human protein found previously to associate with microtubules and the kinesin-interacting neuronal protein DISC1 (Disrupted-in-Schizophrenia 1), but whose cellular function(s) remains unknown. Here we demonstrate that the C. elegans MIP-T3 ortholog DYF-11 is an intraflagellar transport (IFT) protein that plays a critical role in assembling functional kinesin motor-IFT particle compl...
